Starting today, you can build bidirectional communication applications using WebSocket APIs in Amazon API Gateway without having to provision and manage any servers.

HTTP-based APIs use a request/response model with a client sending a request to a service and the service responding synchronously back to the client. WebSocket-based APIs are bidirectional in nature. This means that a client can send messages to a service and services can independently send messages to its clients.

This bidirectional behavior allows for richer types of client/service interactions because services can push data to clients without a client needing to make an explicit request. WebSocket APIs are often used in real-time applications such as chat applications, collaboration platforms, multiplayer games, and financial trading platforms.

This post explains how to build a serverless, real-time chat application using WebSocket API and API Gateway.


SWI stock: How to make thousands in a year after big plunge?

  4. I spent two hours to watch Senate and SolarWind and Microsoft meeting last weekend. 
  5. I made profit over $250 dollars before from Dec. 2020 to Feb. 2021. 
  6. I did over two years network security and cryptograph research, I have strong interest on this area. 
  7. I did study Facebook security officer and his consultation to Solarwind - good learning opportunity;
  8. Feb. 25, 2021, three lessons learned: Do not be too greedy, 4% in two weeks, that is 100% a year return
  9. Second lesson, using trailing stop, when gains drop 20 to 50 dollars, sell them automatically.

I did not know short sale squeeze. Even though I read twitter ex-hedge fund manager post to push EXPR to $13.00 dollars, but I did not believe that it will become true that EXPR stock was pushed to over $13.00 dollars. I could not hold 4800 shares of EXPR and sold over $10.00 dollars. Knowledge is power, and the research I did is not good enough for me to make sound good decision. 

Also I need to think about critical thinking. Every time something his happen, I should consider outside of box, and give it try and assume that there is something I do not know. 

I did equity research but when I sold 4800 shares of EXPR, I did not know wallstreetbet reddit group at all. So my research did not cover most popular group known to millions.

Lessons I learned


Build a good community around myself. I should listen to peers and make quick judgement to take profit. Market is so volatile. It is important for me to survive as a stock investor. 

I should have profited over $10,000 gains on NOK stock, but I did not listen to my friend's advice. I have to keep those NOK stocks 4500 shares and wait for next chance to make profit. 

I should have invested on RYCEY around 10000 shares instead. It is best trustable business in the world and cheapest price in the world.

I understood my weakness. I was not open to consider to purchase those stocks underneath 1 US dollars in 2020. I worried about my 10 years saving to invest on those stocks. I only consider stocks with capital more than 1 billion dollars. I just could not be humble to consider all possible research I can do. 

It is important for me to consider those stocks going up 80 times in one year. Do not chase those hot stocks every one knows called ZOOM or biomedical stocks. I should do my own research and evaluate what are possible if there is a good news about vaccine of pandemic virus.

Introduction

It is challenge for me to explore biotechnology and learn how to invest on biotechnologies. I started to purchase 100 shares of BLUE and 100 shares of CRTX, I like to learn more about vaccine and how to make my investment more fun. 

Vaccine - how to pronounce? 

I did talk to my coworker two months ago, but I could not pronounce Vaccine word correctly. Right now, I started to invest CRTX stock and BLUE stock, OTIC stock and GSK stock. 

I just could not believe that I did not learn anything related to health, biotechnology last 30 years. It is time for me to catch up and make some money. I do believe that I will be very good investor.

IMVT stock: Plan to purchase | study IMVT biotechnology

Next up is Immunovant, a clinical stage biopharmaceutical research firm, focused on developing treatments for patients with autoimmune disorders, a class of diseases in which the immune system attacks the patient’s own body.

The firm’s lead drug candidate, IMVT-1401, is undergoing trials as a treatment for thyroid eye disease, myasthenia gravis, and warm autoimmune hemolytic anemia. The drug described as “a novel, fully human anti-FcRn monoclonal antibody,” delivered by subcutaneous injection.

On February 2, Immunovant’s stock plunged 42%, and it has been falling ever since. The precipitating factor was an announcement by the company that IMVT-1401 has had its Phase 2b clinical trial, for thyroid eye disease, halted temporarily, due to patients experiencing dangerous rises in their LDL levels. LDLs are the potentially harmful form of cholesterol, which have been connected to cardiovascular disease.

Despite the clinical setback, Stiffel’s 5-star analyst Derek Archila reiterated a Buy rating on IMVT shares, along with a $28 price target. This figure suggests a 52% upside potential from current levels. (To watch Archila’s track record, click here)

“Interestingly, increases have only been seen in TED patients, and our review of the literature suggests a few things: (1) it's likely this is TED specific given the biology- see below for details, but we don't think similar LDL increases will be seen in other indications outside TED; and (2) other anti-thyroid therapies used in Graves/TED also see similar increases in LDL, which end up being transient. We think IMVT-1401, in away, is replicating this mechanism," the analyst noted.

Archila summed up, "While we will need to see additional data from the company to confirm... we don't think this program is dead.”

Overall, the Strong Buy analyst consensus view on IMVT would suggest that Wall Street generally agrees with Archila’s assessment. This rating is derived from 8 recent reviews, which include 7 Buys and only a single Hold. The average price target here stands at $40.38, implying ~121% upside for the next 12 months. I like to take value investing approach, buy stocks when it is huge discount. But I have to learn what is going on with CRTX. I waited for less than  a week, and then I had chance to look into the following detail. I just made $380 dollars gains on my two purchases, each time 100 share.

Today, we’re looking at two small-cap biotech firms whose stocks have struck a rut. Each company has hit a recent clinical setback that sent the share price falling, erasing previous gains and sending it back down to low levels.

Setbacks of this sort are not uncommon in the biotech industry, and in fact highlight the risk and speculative nature of the industry.

So what should investors do, when a stock collapses? Is this a matter of poor fundamentals? And has the stock’s price found its low point yet? That’s where the Wall Street pros come in.

Noting that each is set to take back off on an upward trajectory, some 5-star analysts see an attractive entry point for both.

Using TipRanks’ database, we found out that these two tickers have earned Moderate or Strong Buy consensus ratings from the analyst community, and boast strong upside potential.

Cortexyme, Inc. (CRTX)

The first beaten-down name we're looking at is Cortexyme, a clinical-stage biopharma company focused on degenerative diseases, especially Alzheimer’s. The company's lead candidate is COR388, also called atuzaginstat.

Atuzaginstat is currently under investigation in the GAIN trial, a study of its efficacy against Alzheimer’s disease. The trial is fully enrolled, with 643 patients, and the company was moving toward an open label enrollment (OLE) section of the Phase 2/3 study. During a routine regulatory update, Cortexyme announced that the OLE phase would be halted, although the primary GAIN study will continue, with results due to be released in Q4 2021. The announcement of the partial halt triggered a 35% drop in share price.

The partial hold was prompted by adverse events on the liver during the atuzaginstat trial. The hepatic symptoms were reversible and showed no long-term lasting effects. The FDA reviewed these records, and in collaboration with Cortexyme the decision was made to hold the OLE while continuing with GAIN. This decision allows the main thrust of the program to continue, while working out a new protocol for the OLE. The purpose of the OLE is to test long-term efficacy and tolerability of the drug.

In a review of Cortexyme after the announcement, HC Wainwright’s 5-star analyst Andrew Fein noted, “Cortexyme's announcement of a partial clinical hold on the OLE study of atuzaginstat is disappointing, but the reversible nature of the liver toxicity might provide some ray of hope for Cortexyme. We believe that the pivotal trial's continuation suggests that the drug-induced liver injury might not be severe enough to halt the program.”

Turning to the near-term, Fein adds, “Continuation of the GAIN trial is encouraging despite the partial hold on OLE. It suggests that FDA plans to wait for the additional data from the pivotal trial before coming to any conclusion. Management shared that nearly one-third of the GAIN patients have completed the study and way past the 12-week time point, suggesting that they are out of risk.”

To this end, Fein rates CRTX a Buy, and his $76 price target indicates confidence in a 147% growth potential. (To watch Fein’s track record, click here)

Overall, Cortexyme has a Moderate Buy rating from the analyst consensus, with 6 recent reviews breaking down 4 to 1 to 1, Buy-Hold-Sell. The stock’s $83.60 average price target suggests that Wall Street sees a high potential here, on the order of ~170% upside from the trading price of $30.74. (See CRTX stock analysis on TipRanks)

Blue stock: Is There Any Hope for bluebird bio Stock?

I bought 100 shares of BLUE at price $28/ share. This is the first time I had chance to learn more about business. I do think that it is so challenge to innovate without any revenue. It is risky to invest the stock as well. But at least it is honesty to see the future of BLUE. I just could not believe that the price went up above $200/ share, and the stage is III for clinical trial. 

I remember that in FAU from 2001 to 2010 I learned the leukemia as a disease and it is hard to battle against it. 


The share price of bluebird bio (NASDAQ:BLUE) is down a staggering 70% over the past year. The company has encountered more roadblocks than expected on its path to find cures for sickle cell disease (SCD) and beta thalassemia. Worldwide, more than 300,000 babies are born with SCD and over 60,000 are born with beta thalassemia every year.

Patients with SCD have misshapen red blood cells, whereas beta thalassemia patients suffer from a lack of oxygen-carrying hemoglobin in their red blood cells. Patients with these conditions require frequent hospitalization to receive blood transfusions. The only current cure for SCD and beta thalassemia is a bone marrow transplant. Despite trying its best to finding a new cure for these conditions, I don't think it's likely that bluebird bio has the solution.

What happened? 

During a phase 1/2 study involving the company's SCD gene therapy candidate, bb1111, one patient developed acute myeloid leukemia (AML), while there was another reported case of myelodysplastic syndrome (another form of leukemia). The first incident involves a patient treated with bb1111 five years ago who was recently diagnosed with the cancer, while the other was diagnosed with the disease just last week. At the moment, it is not possible to determine with certainty whether or not these serious adverse events are associated with bb1111. There are, however, several factors to consider as a current or potential investor in bluebird bio.

The first is that patients with SCD are twice as likely as those without the disease to develop leukemia. So, it is possible that disease progression itself was the cause of leukemia. However, bb1111's clinical trials involved a total of just 85 participants, so the prevalence of safety issues is far more alarming than if they occurred in a much larger group. 

Second, some evidence suggests that SCD patients who receive hydroxyurea over an extended period of time are also at risk of developing a form of cancer. Hydroxyurea is a generic drug that can help prevent life-threatening episodes of vaso-occlusive crises (VOCs), during which deformed red blood cells in SCD patients accumulate and constrict blood flow. Bluebird enrolled patients in the SCD studies if they failed prior courses of treatment with hydroxyurea, but it also had patients who may not have taken the drug due to lack of tolerance for its side effects.

Lastly, a patient who previously received infusions with bb1111 and subsequently developed AML was cleared of causation last year. However, that study was at least partially funded by bluebird. Scientists are not immune from conflict of interest. When one's research into a safety protocol is funded by the same organization running that protocol, then there exists substantial incentive for the investigator to paint that organization in a more positive light, as opposed to an independent examiner.

That said, it is essential to note that the company is pausing its bb1111 phase 1/2 and phase 3 clinical trials as per protocol, and not necessarily because bb1111 is leukemogenic -- or, because it can cause leukemia. When significant, unexpected safety issues arise (such as patients developing cancer), investigators must pause the trial to determine whether or not the adverse events are related to the experimental therapy. It can be relative common or very rare for this happen during the clinical trial process, depending on the number of participants involved. 

Can bluebird bio survive this? 

Bluebird's gene therapies have somewhat proven themselves in previous clinical studies. bb1111 completely eliminated the occurrence of VOCs after infusion in SCD patients. Another genetic treatment candidate, Zynteglo, relieved 90% of beta thalassemia patients of blood transfusions post treatment. It has received Condition Marketing Authorization from the European Medical Agency. However, the U.K.'s regulatory body, the National Institute for Health and Care Excellence (NICE), rejected Zynteglo. NICE cites the small number of trial participants and the lack of long-term follow-up as reasons for its decision. Previously, bluebird had expected that it could charge at least 1.45 million pounds for a one-time infusion of the therapy. In response to recent patient health developments, bluebird has stopped marketing Zynteglo in Europe for the time being.

Right now, bluebird is spending over $450 million per year on research and development, with no product revenue. Due to the recent serious adverse events, it will probably fall short of filing a Biologic License Application for bb1111 and launching Zynteglo in the U.S. by 2022.  

Its market cap of $1.7 billion is incredibly low, much lower than Global Blood Therapeutics' (NASDAQ:GBT) $3.2 billion. Global Blood Therapeutics is a company that markets a marginally effective treatment for SCD, Oxbryta. (Oxbryta is a medication that slightly improves the hemoglobin levels of patients with SCD, but has not been shown to actually reduce the number of VOCs that occur.) But bluebird's market cap is down and its stock is cheap for good reason. It is already down nearly 70% from where it was a year ago.

The reality is that we do not now whether bb1111 causes leukemia, and we probably won't know until the investigation into the trial finishes. The company could well see its fortunes reverse should Zynteglo or bb1111 be administered to a larger number of participants and followed-up on for extended durations. For these reasons, I would recommend investors avoid the stock until it can prove that bb1111 is safe and Zynteglo can reproduce its results in more extensive studies, which could take years. In the meantime, there are plenty of other fantastic biotechs with safer drug lineups to add to your portfolio.
